AOP Developer's Handbook 2.5: Supplement to the Guidance Document for Developing and Assessing AOPs

Impact/Purpose:

This document provides an update to the AOP Developer's Handbook (formerly AOP Users' Handbook). The handbook serves as the primary "instructions for authors" that guides the development of a formal adverse outcome pathway (AOP) description in the AOP-Wiki (aopwiki.org).  The current version implements key updates to the AOP framework including: (1) collection of information on the AOP development strategy and evidence assembly strategy employed by the authors; (2) efforts to reduce conflation of AOP development with stressor-specific hazard assessment; (3) increased visibility of information related to modulating factors; (4) minor updates to the AOP naming convention; and (5) improved examples of table formats for collecting and evaluating weight of evidence in association with AOP development. Updates to the handbook are synced with software updates implementing corresponding features in the AOP-Wiki system. This represents the first major updates to the guidance on AOP development since 2018. Guidance implemented in the current version of the Handbook is expected to increase the utility of AOPs to support the application of new approach methodologies in chemical safety-related decision-making.

Description:

AOP-Wiki release 2.5 is intended to implement the following feature upgrades/changes to the AOP-Wiki (aopwiki.org):  (1) automatic assignment of molecular initiating event (MIE) and adverse outcome (AO) designations in the AOP search table; (2) ability to filter displayed AOPs by OECD status; (3) incorporation of link-outs to 3rd party tools that utilize AOP-Wiki information; (4) implementation of an electronic users handbook and help upgrades; (5) addition of new text fields for adding details on AOP development and evidence collection strategies;
